Public transportation will get you there. Take AirTrain from terminal to Jamaica Station ($5), transfer to LIRR ($8.50/$6.25) or E subway ($2.25) to Penn Station, transfer to Amtrak (faster but more expensive-$40-$60) or NJ Transit NE Corridor train ($15.50) to Trenton, transfer to the Septa regional train ($3.75) towards Center City-Philly and get off at Bristol. Take the 129 bus ($2.50) towards Oxford Valley Mall near the hotel. You could also try a shared ride shuttle from JFK, inquire at Ground Transportation.

I am taking a bus into Port Authority. What is the easiest way to get to JFK? Thanks.

NY Airporter bus from PA to JFK is $15.50 or take Jamaica-bound E subway ($2.25) from PA to Sutphin Blvd/JFK and transfer to AirTrain ($5).

I need to get from JFK to the Nassau Blvd Station to visit Adelphi College. Do I take the JFK Airtrain Station to Jamaica Station and then Jamaica to Nassau Boulevard? Or should I just take a cab?

Taxi is most direct and would cost around $45. You can certainly take LIRR-Hempstead Line from Jamaica to Nassau Blvd. ($6.75 peak/$5 off-peak) and local taxi or a short walk to the campus.

I need to go from JFK to Hampton Inn Madison Square Garden, what is the best way to go?

Take AirTrain from terminal ($5) to Jamaica Station, transfer to either E subway ($2.25) or LIRR ($8.50 peak/$6.25 off peak) to Penn Station. Exit to 7th Ave, walk two blocks south to 31st St and turn left, hotel is between 6th & 7th Aves.

I need to go from JFK to the United Nations Building early morning. which subway or LIRR?

Thanks

The closest subway to the UN is at 42nd St/Grand Central and walk four long blocks east on 42nd St to the UN. Take AirTrain from terminal to Jamaica Station, transfer to E subway to Lexington Ave/53rd St, transfer to downtown #6 subway one stop to 42nd St. You can save the walk by taking the eastbound M42 crosstown bus from subway to 1st Ave.
